Western blot of CLASP2 in mouse brain (lanes 1 & 3) and rat PC12 cells (lanes 2 & 4). The blots were probed with rat monoclonals CM5051 anti-CLASP2 (clone KT68) (lanes 1 & 2) and CM5071 anti-CLASP2 (clone KT69) (lanes 3 & 4). The antibodies were used at a dilution of 1:500.

Immunocytochemical labeling of CLASP2 in paraformaldehyde-fixed and NP40-permeabilized A431 cells. The cells were dual labeled with mouse monoclonal anti-α-Tubulin (TM4111) (left) and rat monoclonal anti-CLASP2 (CM5051) (middle). The antibodies were detected using either goat anti-mouse:DyLight® 488 (MS3011) or goat anti-Rat:DyLight® 594 (RS3111).

Background
The microtubule (MT) plus-end is a crucial site for the regulation of MT dynamics and interactions by several groups of plus-end tracking proteins (+TIPs). These +TIPs form comet-like accumulations at the plus ends of MTs to regulate MT dynamics and interactions with organelles and macromolecular complexes. The +TIPs include diverse groups of proteins, such as motor and nonmotor proteins, MT polymerases and depolymerases as well as various regulatory and adaptor proteins. The CLIP-associated protein (CLASP) family includes CLASP1 and CLASP2 proteins, which are expressed as long (α) and short (β) isoforms. Thse +TIPs conatin an N-terminal TOG domain, multiple TOG-like domains, and a basic and serine-rich motif (SxIP). The TOG domain facilitates interaction with tubulin dimers, while the SxIP motif promotes interaciton with EB1 and MTs. A C-terminal domain is involved in interaction with CLIPs, as well as several other proteins. CLASPs are MT stabilizing fators that localize to mitotic spindles, kinetochores, and the midbody. CLASPs are important for cell division, and may regulate cell migration and neuronal growth cone motility.

Immunogen
Clone KT68 was generated from a GST fusion protein containing the N-terminus of mouse CLASP2. This sequence is highly conserved in human and rat CLASP2, and has low homology to CLASP1.
Buffer and Storage
Rat monoclonal antibody purified with protein G chromatography is supplied in 100µl phosphate-buffered saline and 0.05% sodium azide. Aliquot and Store at –20°C. Stable for 1 year.
Specificity
The antibody detects a 160 kDa* doublet corresponding to the molecular mass of CLASP2 on SDS-PAGE immunoblots of human HeLa, mouse brain, and rat PC12 cells. The antibody also detects CLASP2 by immunocytochemistry in HeLa and A431 cells.

*All molecular weights (MW) are confirmed by comparison to Bio-Rad Rainbow Markers and to western blot mobilities of known proteins with similar MW.
